Background:

  • The Titanium-44 (44Ti)/Scandium-44 (44Sc) generator offers cyclotron-independent production of 44Sc.
  • 44Sc is a positron-emitting radionuclide valuable for Positron Emission Tomography (PET) imaging.

Purpose of the Study:

  • To develop a post-elution processing method for 44Sc generator eluates.
  • The aim is to reduce eluate volume, decrease HCl concentration, and remove oxalate anions.
  • To enhance the purity of 44Sc for radiolabeling applications.

Main Methods:

  • On-line adsorption of 44Sc onto a cationic resin (AG 50W-X8, H+-form).
  • Desorption of purified 44Sc using ammonium acetate solution (0.25M, pH 4.0).
  • Evaluation of processing time, yield, and reduction of Titanium-44 (44Ti) breakthrough.

Main Results:

  • Achieved >98% efficacy for 44Sc adsorption onto the cationic resin.
  • The post-processing procedure takes approximately 10 minutes.
  • Overall yield of purified 44Sc reached 90% relative to the generator output.
  • Reduced 44Ti breakthrough by one order of magnitude.
  • Delivered 150MBq of 44Sc with <10Bq of 44Ti from an initial 185MBq generator.

Conclusions:

  • The developed post-elution processing method is efficient and rapid for purifying 44Sc.
  • This technique significantly improves 44Sc purity by reducing volume, HCl, and 44Ti contaminants.
  • The purified 44Sc is suitable for immediate use in radiolabeling for PET imaging.
