Summary

Researchers identified a novel regulatory T cell (Treg) population, CD4(+)CD25(-)LAG-3(+) Tregs, in mice. These Tregs express early growth response gene 2 (Egr-2) and IL-10, demonstrating immunosuppressive functions and a potential human counterpart.

Area of Science:

  • Immunology
  • Cell Biology
  • Molecular Biology

Background:

  • A novel murine regulatory T cell (Treg) population, CD4(+)CD25(-)LAG-3(+) Tregs, was previously identified.
  • These Tregs exhibit a distinct mRNA expression profile compared to conventional CD4(+)CD25(+) Tregs.

Purpose of the Study:

  • To investigate the characteristics of murine CD4(+)CD25(-)LAG-3(+) Tregs.
  • To identify a potential human counterpart of these murine Tregs.

Main Methods:

  • Microarray analysis to compare gene expression profiles.
  • Quantitative PCR to assess IL-10 expression.
  • Retroviral gene transfer to study the role of Egr-2.
  • In vitro and in vivo functional assays to evaluate immunosuppressive activity.

Main Results:

  • Murine CD4(+)CD25(-)LAG-3(+) Tregs uniquely express early growth response gene 2 (Egr-2), associated with anergy induction.
  • These Tregs produce high levels of IL-10 upon T cell receptor stimulation and exhibit immunosuppressive activity.
  • Egr-2 gene transfer into naive CD4(+) T cells induced IL-10 secretion, LAG-3 expression, and antigen-specific immunosuppression in vivo.
  • Human CD4(+)CD25(-)LAG-3(+) T cells were identified with similar patterns to murine Tregs, expressing high IL-10 levels and showing comparable gene expression profiles.

Conclusions:

  • Murine CD4(+)CD25(-)LAG-3(+) Tregs possess unique molecular and functional characteristics, including Egr-2 expression and IL-10 production.
  • Egr-2 plays a crucial role in inducing immunosuppressive functions in CD4(+) T cells.
  • Human CD4(+)CD25(-)LAG-3(+) T cells represent a potential human counterpart to murine CD4(+)CD25(-)LAG-3(+) Tregs, suggesting conserved regulatory mechanisms.
