Summary

DNA profiles were successfully obtained from used immunochromatographic test devices, even after three months of room temperature storage. This demonstrates the potential for DNA recovery from these forensic evidence sources.

Area of Science:

  • Forensic Science
  • Molecular Biology
  • Genetics

Background:

  • Immunochromatographic test devices are used for rapid identification of biological fluids.
  • The stability of DNA on these devices after use and storage is crucial for forensic investigations.
  • Previous research has not extensively explored DNA profiling from aged test devices.

Purpose of the Study:

  • To determine if DNA profiles can be successfully retrieved from used immunochromatographic test devices.
  • To assess the impact of storage duration at room temperature on DNA recovery.
  • To validate the obtained DNA profiles against reference samples.

Main Methods:

  • Used immunochromatographic cards (blood and saliva) were stored at room temperature for varying durations.
  • DNA was extracted using a low copy number (LCN) procedure.
  • Autosomal and Y-STR DNA amplification was performed using specific PCR kits.
  • DNA profiles were analyzed via capillary electrophoresis and specialized software.

Main Results:

  • DNA profiles were obtained from most cards stored for up to three months.
  • Successful DNA profiling was achieved from all four types of immunochromatographic cards tested.
  • Generated DNA profiles were concordant with the donors' reference samples.

Conclusions:

  • Immunochromatographic test devices can serve as a source for reliable DNA profiling.
  • DNA remains recoverable from these devices after extended storage periods at room temperature.
  • This finding has significant implications for forensic casework involving biological evidence.
