GPCRs Regulate Adenylyl Cylase Activity01:09

GPCRs Regulate Adenylyl Cylase Activity

Some GPCRs transmit signals through adenylyl cyclase (AC), a transmembrane enzyme. AC helps synthesize second messenger cyclic adenosine monophosphate (cAMP). AC catalyzes cyclization reaction and converts ATP to cAMP by releasing a pyrophosphate. The pyrophosphate is further hydrolyzed to phosphate by the enzyme pyrophosphatase, which drives cAMP synthesis to completion. However, cAMP is rapidly degraded to 5′ AMP by the enzymes phosphodiesterase (PDE), preventing overstimulation of cells.

Glucagon-like Receptor Agonists01:24

Glucagon-like Receptor Agonists

Incretins include glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP), which stimulate insulin secretion post-meals. In type 2 diabetes, GIP's efficacy is reduced, making GLP-1 a viable drug target. GIP originates from preproGIP.
GLP-1, when administered in high doses intravenously, triggers insulin secretion, inhibits glucagon release, slows gastric emptying, reduces food intake, and restores normal insulin secretion. However, its rapid inactivation by the...
Hormones of the Adrenal Glands01:31

Hormones of the Adrenal Glands

Adrenal hormones play a pivotal role in maintaining the body's electrolyte balance and orchestrating responses to stress, showcasing the intricate functions of the adrenal cortex and medulla.
The adrenal cortex, a powerhouse of hormone synthesis, generates over two dozen corticosteroid hormones. The zona glomerulosa produces mineralocorticoids, exemplified by aldosterone, influencing the electrolyte composition of body fluids. The synthesis of glucocorticoids such as cortisol and corticosterone...
Cushing Syndrome II: Pathophysiology01:19

Cushing Syndrome II: Pathophysiology

Cortisol production is normally governed by the hypothalamic–pituitary–adrenal (HPA) axis, which maintains hormonal balance through tightly regulated feedback mechanisms. Disruption of this regulatory system is central to the development of Cushing syndrome, whether the excess cortisol originates from external medications or internal pathology. Persistent cortisol elevation alters metabolism, immune function, and endocrine signaling, producing the characteristic clinical features of the...

Summary

Glucocorticoids (GC) and high-fat diets reduce serum adiponectin levels in rats. This decrease is linked to inhibited adiponectin mRNA expression in white adipose tissue (WAT), indicating a synergistic effect.

Area of Science:

  • Endocrinology
  • Metabolic Syndrome Research

Background:

  • Adiponectin is crucial in regulating obesity and insulin resistance.
  • Glucocorticoids (GC) are known to inhibit adiponectin expression in vitro, but in vivo effects are debated.

Purpose of the Study:

  • To investigate the in vivo effect of GC on serum adiponectin levels.
  • To examine the impact of GC on adiponectin expression in white adipose tissue (WAT).
  • To assess the combined effect of high-fat diet and GC on adiponectin levels in rats.

Main Methods:

  • Obese rat models were established using a high-fat diet.
  • Rats (non-obese and obese) received daily intraperitoneal injections of normal saline, low-dose GC, or high-dose GC for 20 days.
  • Serum adiponectin was quantified using ELISA, and adiponectin mRNA levels were analyzed via Northern blot.

Main Results:

  • An 80-day high-fat diet significantly decreased serum adiponectin levels in rats.
  • A 20-day GC injection period significantly reduced serum adiponectin in both non-obese and obese rats.
  • High-dose GC administration notably decreased adiponectin mRNA expression in epididymal WAT of both rat groups.

Conclusions:

  • High-fat diet consumption leads to reduced serum adiponectin levels in rats.
  • Glucocorticoids significantly lower serum adiponectin, potentially by inhibiting adiponectin mRNA expression in WAT.
  • High-fat diet and glucocorticoids exhibit a synergistic inhibitory effect on adiponectin expression in rats.
