Genetic variations in IL-1 cluster genes are not linked to subarachnoid hemorrhage susceptibility. However, the IL-1beta gene may influence disease severity in patients with intracranial aneurysms.
Area of Science:
- Neuroscience
- Genetics
- Immunology
Background:
- Proinflammatory cytokines, like Interleukin-1 (IL-1), are implicated in intracranial aneurysm pathogenesis.
- IL-1 plays a key role in central nervous system injuries.
Purpose of the Study:
- To examine the association between polymorphisms in IL-1alpha, IL-1beta, and IL-1 receptor antagonist genes and susceptibility to subarachnoid hemorrhage (SAH) from ruptured intracranial aneurysms.
- To investigate the relationship between these gene polymorphisms and clinical characteristics of SAH.
Main Methods:
- Genotyping of IL-1alpha (-889), IL-1beta (-511), and IL-1 receptor antagonist (VNTR) genes in 215 SAH patients and 155 controls.
- Analysis of allelic and genotypic frequencies.
- Review of patient files for clinical data at admission and 6-month follow-up.
Main Results:
- No significant association was found between the examined IL-1 gene polymorphisms and susceptibility to aneurysmal SAH.
- Patients with the IL-1beta (T/T) genotype showed higher Hunt and Hess scores at admission (P = .034) and poorer 6-month Glasgow Outcome Scale scores (P = .026).
- No other polymorphisms affected clinical features.
Conclusions:
- Genetic variations in the studied IL-1 cluster genes are not associated with aneurysmal SAH susceptibility.
- The IL-1beta gene polymorphism may influence the severity of aneurysmal SAH and could be considered a disease severity gene.
