Hybridoma Technology01:31

Hybridoma Technology

Hybridoma technology is used for the large-scale production of monoclonal antibodies. Monoclonal antibodies bind to only a single antigenic determinant or epitope. Such antibodies are used in research, diagnostics, and disease therapy. The hybridoma technology established in 1975 by Georges Köhler and Cesar Milstein was awarded the Nobel Prize in Medicine in 1984 for revolutionizing research and therapy.

Area of Science:

  • Immunology
  • Biotechnology

Background:

  • Human monoclonal antibodies (mAbs) are increasingly vital therapeutics for various human diseases.
  • Human antibody repertoires are a key resource for generating these therapeutic mAbs.

Purpose of the Study:

  • To review natural and non-natural human antibody repertoires.
  • To discuss strategies for mining these repertoires to generate human mAbs.
  • To highlight methods for selecting human mAbs with optimal properties.

Main Methods:

  • Exploration of non-combinatorial and combinatorial repertoire mining strategies.
  • Discussion of selection methods including hybridoma technology, B cell expansion, single-cell PCR, and display technologies (phage, yeast, mammalian cell).
  • Analysis of different repertoire types: naïve, immune, transgenic, and synthetic.

Main Results:

  • Various strategies exist for mining human antibody repertoires.
  • Selection methods vary in efficiency and applicability to different repertoire types.
  • Advancements in methods are enabling the generation of human mAbs with enhanced pharmacokinetic and pharmacodynamic profiles.

Conclusions:

  • The choice of strategy for human mAb generation depends on repertoire characteristics and desired therapeutic properties.
  • Ongoing refinement of mining and selection methods is crucial for efficient development of superior human mAbs.
  • Human antibody repertoires offer a rich platform for the discovery and development of next-generation therapeutics.
