The Thyroid Gland

The thyroid gland is a small, butterfly-shaped gland located in the neck and covers the anterior surface of the trachea. The gland has two lateral lobes connected by a thin tissue mass called the isthmus. Internally, each lobe comprises many small spherical structures known as thyroid follicles, surrounded by a network of blood vessels.

Summary

Quantitative RT-PCR analysis of UbcH10 mRNA shows promise in diagnosing thyroid fine-needle aspiration (FNA) samples. This method, unlike immunohistochemistry, can help differentiate benign from malignant thyroid lesions.

Background:

  • Thyroid fine-needle aspiration (FNA) diagnosis faces challenges with follicular neoplasm/suspicious for malignancy classifications.
  • UbcH10 has emerged as a potential marker for various neoplasms, including thyroid cancer.

Purpose of the Study:

  • To evaluate the diagnostic utility of UbcH10 expression in thyroid FNAs using quantitative reverse transcriptase polymerase chain reaction (RT-PCR) and immunohistochemistry.
  • To compare UbcH10's diagnostic accuracy with existing markers and gene assays.

Main Methods:

  • Analysis of 84 thyroid FNAs with histological follow-up (30 malignant).
  • UbcH10 and Ki-67 expression assessed by immunohistochemistry.
  • UbcH10, CCND2, and PCSK2 mRNA levels quantified using RT-PCR.
  • Diagnostic accuracy determined by receiver operating characteristic (ROC) curve analysis (Area Under the Curve - AUC).

Main Results:

  • Immunohistochemistry for UbcH10 showed limited diagnostic value due to sporadic staining and a low cutoff value.

  • Quantitative RT-PCR analysis of UbcH10 mRNA is a valuable tool for improving malignancy detection in thyroid FNAs.
  • UbcH10 can be incorporated into RT-PCR-based diagnostic panels for thyroid cancer.
  • Immunohistochemistry for UbcH10 is not recommended for routine diagnostic application in these cases.
