    Purpose:

    • To compare the efficacy and toxicity of contemporary radiotherapy versus combined treatment including radioprotector GGS-9 and 3D conformal radiotherapy (3D CRT) for prostate cancer.

    Summary:

    • Contemporary radiotherapy (n=37) involved total tumor doses (TTD) up to 70 Gy. Combined treatment (n=51) included GGS-9 and 3D CRT, with TTD up to 76 Gy.
    • GGS-9 reduced acute radiation injury from 56.7% to 11.7% (p=0.0001). 3D CRT lowered late-onset injury from 13.5% to 3.9% (p=0.01).
    • Recurrence-free survival improved from 73% with contemporary radiotherapy to 94.2% with 3D CRT (p=0.0001).

    Impact:

    • Combined radiotherapy with GGS-9 and 3D CRT significantly reduces acute and late radiation injuries in prostate cancer patients.
    • Improved dose distribution with 3D CRT allows for higher tumor doses without increasing toxicity.
    • This approach enhances local control and significantly improves 5-year recurrence-free survival rates.
