Purpose of the Study:

  • To investigate aripiprazole's effects on established MAP-induced behavioral sensitization in mice.
  • To explore the receptor mechanisms underlying aripiprazole's action.

Main Methods:

  • Mice received MAP (1.0mg/kg) for 10 days to induce sensitization.
  • Aripiprazole was administered during withdrawal from MAP.
  • Locomotor activity was measured after a MAP challenge dose.
  • Effects of sulpiride, risperidone, and WAY100635 were also assessed.

Main Results:

  • Established MAP-induced behavioral sensitization was attenuated by repeated aripiprazole treatment during withdrawal.
  • Sulpiride and risperidone did not replicate aripiprazole's effect.
  • Aripiprazole's attenuation was blocked by the 5-HT1A antagonist WAY100635.

Conclusions:

  • Aripiprazole's attenuation of MAP-induced sensitization is mediated by serotonin 5-HT1A receptors.
  • Aripiprazole may hold therapeutic value for drug-induced psychosis and schizophrenia.
