Summary

This study examined how ceramides and cholesterol in healthy skin change with age and sex. Researchers collected skin samples from 55 people with no skin conditions and measured ceramide subgroups and ceramide/cholesterol ratios. They also looked at how these levels relate to dry skin and age. The results showed no significant differences in ceramides between young and old skin. However, men had higher ceramide/cholesterol ratios than women. The study also found no link between self-reported dry skin and ceramide levels. These findings suggest that ceramide composition in healthy skin is stable across age groups and may vary by sex.

Background:

It was already known that lipids in the outermost layer of the skin play a central role in maintaining skin barrier function. However, no prior work had resolved how these lipid compositions vary among individuals with healthy skin. While eczematous conditions are associated with lipid imbalances, limited data exist on lipid composition in healthy skin. Researchers propose that age, sex, and subjective dryness may influence ceramide subgroups and ceramide/cholesterol ratios. This gap motivated the investigation of lipid profiles in healthy skin across different demographics. No prior work had resolved whether age or sex correlates with ceramide levels in healthy individuals. Prior research has shown that ceramides are essential for epidermal barrier integrity. That uncertainty drove the need to examine ceramide subgroups and their ratios in a healthy population. No prior work had resolved whether self-reported dryness correlates with ceramide levels in healthy skin.

Purpose Of The Study:

The aim of the study was to compare ceramide subgroups and ceramide/cholesterol ratios in healthy skin across age and sex groups. The researchers sought to determine whether ceramide profiles differ in young versus old skin. They also investigated whether ceramide levels correlate with subjective dryness in a subset of participants. This study aimed to address the lack of data on lipid variations in healthy skin. The purpose was to examine whether age or sex influences ceramide composition in the stratum corneum. The researchers wanted to assess whether self-reported dryness correlates with ceramide levels. The study aimed to evaluate ceramide/cholesterol ratios in male and female skin. The purpose was to provide baseline data on lipid composition in healthy skin across demographics.

Main Methods:

The study included 55 participants with healthy skin and no history of eczema. Researchers collected stratum corneum lipid profiles using appropriate extraction techniques. Transepidermal water loss was measured as an indicator of barrier function. A questionnaire was used to assess subjective dryness in a subset of 16 participants.
