Summary

Apolipoprotein E (apoE) levels in the brain are highest in young mice, decreasing by adulthood and remaining stable in old age. This suggests other factors maintain brain function during aging.

Background:

  • Apolipoprotein E (apoE) is a key lipid transport protein crucial for brain membrane repair and synaptic plasticity.
  • ApoE genotype is linked to Alzheimer's disease (AD) risk, and its expression elevates following nervous system injury and in age-related disorders like AD.
  • The precise changes in apoE levels during normal brain aging remain unclear.

Purpose of the Study:

  • To investigate the impact of normal aging on apoE protein and mRNA levels in the mouse cerebral cortex.
  • To determine how apoE expression changes across different age groups: young, adult, and old.

Main Methods:

  • Analysis of apoE protein levels using Western blotting.
  • Quantification of apoE mRNA levels via Northern blotting and RT-PCR.
  • Comparison of apoE expression in the cerebral cortex of male AKR strain mice at young, adult, and old ages.
