Background:

  • Ozone (O3) is a major air pollutant.
  • Ozone exposure can induce oxidative stress in the lungs.
  • Antioxidant enzymes (AOEs) play a crucial role in cellular defense against oxidative damage.

Purpose of the Study:

  • To investigate the effect of ozone exposure on antioxidant enzyme activity and gene expression in rat lungs.
  • To determine the pre-translational mechanisms underlying ozone-induced changes in AOE expression.

Main Methods:

  • Rats of varying ages were exposed to 0.7 ppm ozone for 1-5 days.
  • Lung homogenates were analyzed for AOE activity (Cu,Zn superoxide dismutase, Mn SOD, catalase, glutathione peroxidase).
  • mRNA concentrations for AOEs were quantified using molecular techniques.

Main Results:

  • Ozone exposure significantly increased the activity of multiple AOEs in rat lungs after 5 days.
  • Elevated mRNA concentrations for AOEs were observed, correlating with increased enzyme activity.
  • mRNA levels increased by day 3 of ozone exposure in adult rats.
  • Differences in mRNA concentration were more pronounced than differences in enzyme activity.

Conclusions:

  • Ozone exposure upregulates the expression of antioxidant enzyme genes in rat lungs.
  • The increased expression is likely mediated pre-translationally, primarily at the transcriptional level.
  • This suggests a coordinated transcriptional response to ozone-induced oxidative stress.
