Type II Diabetes II: Pathophysiology01:24

Type II Diabetes II: Pathophysiology

PathophysiologyType 2 diabetes mellitus (T2DM ) is a chronic metabolic disorder characterized by insulin resistance and progressive pancreatic β-cell dysfunction, leading to impaired glucose homeostasis. It results from interactions among genetic predisposition, environmental factors, and metabolic stressors, such as overnutrition and a sedentary lifestyle.Insulin Resistance and Glucose DysregulationEarly T2DM involves insulin resistance in skeletal muscle, adipose tissue, and the liver.
Cell Specific Gene Expression01:58

Cell Specific Gene Expression

Multicellular organisms contain a variety of structurally and functionally distinct cell types, but the DNA in all the cells originated from the same parent cells. The differences in the cells can be attributed to the differential gene expression. Liver cells, whose functions include detoxification of blood, production of bile to metabolize fats, and synthesis of proteins essential for metabolism, must express a specific set of genes to perform their functions. Gene expression also varies with...
Hormones Regulating Blood Glucose01:16

Hormones Regulating Blood Glucose

Insulin is released by beta cells of the pancreas when blood glucose levels are high. It facilitates glucose absorption and utilization in insulin-dependent cells with insulin receptors on their plasma membranes. Insulin promotes glucose uptake by increasing the number of glucose transport proteins in the cell membrane, allowing glucose to enter the cell. As a result, glucose utilization and ATP production are enhanced.
In addition to accelerating glucose uptake and utilization, insulin has...
Diabetic Retinopathy01:27

Diabetic Retinopathy

DefinitionDiabetic retinopathy is a microvascular complication of diabetes affecting the retinal blood vessels.Risk FactorsDiabetic retinopathy is present in almost all individuals with type 1 diabetes and more than 60% of those with type 2 diabetes after two decades of disease. The risk increases with poor glycemic control, hypertension, dyslipidemia, smoking, pregnancy, and puberty.Although cataracts and glaucoma are also more frequent in people with diabetes, retinopathy remains the leading...

Summary

Circulating pigment epithelium-derived factor (PEDF) is linked to insulin resistance in men. Weight loss significantly reduces PEDF levels, suggesting its role in adipocyte biology and metabolic health.

Area of Science:

  • Metabolic research
  • Endocrinology
  • Adipocyte biology

Background:

  • Pigment epithelium-derived factor (PEDF) is a protein with diverse biological functions.
  • Its role in human metabolic diseases, particularly insulin resistance, requires further elucidation.
  • Understanding PEDF's involvement in adipocyte function is crucial for metabolic health research.

Purpose of the Study:

  • To investigate the association between circulating PEDF levels and insulin resistance in vivo.
  • To examine PEDF expression and secretion in human adipocytes in vitro.
  • To determine the effect of weight loss on circulating PEDF concentrations.

Main Methods:

  • Circulating PEDF levels were measured using ELISA in 125 Caucasian men.
  • Metabolic profiles, including insulin sensitivity and glucose tolerance, were assessed.
  • PEDF gene expression and secretion were analyzed during human preadipocyte differentiation.

Main Results:

  • Circulating PEDF positively correlated with BMI, waist-to-hip ratio, HbA1c, and triglycerides, and negatively with insulin sensitivity.
  • PEDF levels were elevated in individuals with impaired glucose tolerance and type 2 diabetes.
  • Weight loss significantly decreased PEDF concentrations, which was associated with improvements in obesity, insulin resistance, and blood pressure.

Conclusions:

  • Circulating PEDF is significantly associated with insulin sensitivity in humans.
  • For the first time, this study demonstrates a significant decrease in PEDF concentrations following weight loss.
  • The findings suggest PEDF plays a role in human adipocyte biology and metabolic regulation.
