Super-resolution Fluorescence Microscopy01:37

Super-resolution Fluorescence Microscopy

Super-resolution fluorescence microscopy (SRFM) provides a better resolution than conventional fluorescence microscopy by reducing the point spread function (PSF). PSF is the light intensity distribution from a point that causes it to appear blurred. Due to PSF, each fluorescing point appears bigger than its actual size, and it is the PSF interference of nearby fluorophores that causes the blurred image. Various approaches to achieving higher resolution through SRFM have recently been developed.

Area of Science:

  • Biomedical Optics
  • Microscopy
  • Biophotonics

Background:

  • Optical biopsy offers a non-invasive alternative to traditional tissue sampling.
  • Existing fiber-microscopy techniques often lack the resolution or speed for real-time in vivo applications.

Purpose of the Study:

  • To develop a miniaturized, high-resolution, video-rate fiber-microscope for in vivo optical biopsy of human skin.
  • To evaluate the performance of a novel epi-third-harmonic-generation (epi-THG) system for skin imaging.

Main Methods:

  • Development of a miniaturized epi-third-harmonic-generation (epi-THG) fiber-microscope utilizing a micro-electro-mechanical system (MEMS) mirror.
  • Integration of a large-mode-area (LMA) photonic crystal fiber (PCF) and a microscopic objective to minimize pulse distortion.
  • Achieving a lateral resolution of 0.4 micrometers for epi-THG signals.

Main Results:

  • Successful development of a compact fiber-microscope with a video frame rate of 31 Hz.
  • Demonstration of in vivo, real-time virtual biopsy of Asian skin with sub-micron resolution.
  • Significant reduction in nonlinear distortion of ultrafast pulse delivery.

Conclusions:

  • The developed miniaturized epi-THG fiber-microscope is suitable for least invasive, hand-held clinical use.
  • The system provides high-resolution, real-time imaging capabilities for optical biopsy.
  • This technology has the potential to advance non-invasive dermatological diagnostics.
