Background:

2,2',4,4'-Tetrabromodiphenyl ether (BDE-47) is a flame retardant with increasing environmental and human presence.
Thyroid disruption is a sensitive health effect linked to BDE-47 exposure.

Purpose of the Study:

To investigate the interaction between BDE-47 and the thyroid receptor beta (TRβ).
To determine if BDE-47 directly affects TRβ binding or transcriptional activity.

BDE-47 did not compete with [(125)I]T(3) for TRβ binding, even at high concentrations.
BDE-47 did not affect TRβ-mediated transcriptional regulation in vitro.
Differential expression of thyroid responsive genes was observed in the liver and brain of exposed rat pups, independent of TRβ activity.

Conclusions:

BDE-47 does not directly interact with TRβ1 or influence its transcriptional activity.
Developmental exposure to BDE-47 causes differential gene expression in the liver and brain through an unelucidated mechanism.
