Heat acclimation establishes a long-lasting cellular memory through epigenetic changes in histone modifications. This histone acetylation primes cells for faster heat stress response and protection.
Area of Science:
- Physiology
- Molecular Biology
- Epigenetics
Background:
- Heat acclimation (AC) induces lasting transcriptional changes in cytoprotective genes, suggesting epigenetic memory.
- This memory may involve upstream epigenetic information facilitating rapid reacclimation (ReAC) and cytoprotection.
Purpose of the Study:
- To investigate the role of posttranslational histone modifications in heat acclimation memory.
- To determine if histone modifications link to rapid reacclimation and cytoprotective memory.
Main Methods:
- Rats underwent heat acclimation (AC), deacclimation (DeAC), and reacclimation (ReAC) protocols.
- Examined histone H4 acetylation and histone H3 acetylation/phosphorylation at heat shock element (HSE) of hsp70 and hsp90 promoters.
- Assessed TIP60 recruitment and Heat Shock Factor-1 (HSF-1) binding to HSE.
Main Results:
- AC elevated histone H4 acetylation at HSE of hsp70 and hsp90 promoters.
- Histone H3Ser10 phosphorylation occurred during early AC (2-day).
- HSF-1 binding to hsp70 HSE was sustained throughout AC-DeAC-ReAC; hsp90 showed transient loss of binding during DeAC, recovering upon ReAC, correlating with HSP-90 protein levels.
Conclusions:
- Early histone H3 phosphorylation may initiate subsequent histone H4 acetylation.
- Constitutively acetylated histone H4 and maintained euchromatin state contribute to rapid cytoprotective acclimation memory.
- Epigenetic modifications, particularly histone acetylation, are crucial for cellular memory of heat acclimation.
