Summary

The opto-fluidic ring resonator (OFRR) biosensor detects DNA methylation. MBD-2 protein and antibodies were compared for capturing methylated DNA, with MBD-2 showing better binding to double-stranded DNA.

Area of Science:

  • Biomedical Engineering
  • Molecular Biology
  • Biosensing Technology

Background:

  • Opto-fluidic ring resonators (OFRR) are sensitive, label-free optical biosensors.
  • DNA methylation analysis is crucial for cancer research and clinical diagnostics.
  • Methyl binding domain (MBD) proteins are key in recognizing methylated DNA.

Purpose of the Study:

  • To evaluate the opto-fluidic ring resonator (OFRR) for label-free analysis of oligonucleotide methylation.
  • To investigate the use of MBD-2 protein as a capture molecule for methylated DNA.
  • To compare the binding affinities of MBD-2 and an anti-5-methylcytidine antibody for methylated DNA.

Main Methods:

  • Quantitative characterization of OFRR response to methylated single-stranded DNA (ssDNA) and double-stranded DNA (dsDNA).
  • Assessment of OFRR performance based on the number of methylated cytosines and DNA concentration.
  • Comparative analysis of MBD-2 protein versus anti-5-methylcytidine antibody as capture agents.

Main Results:

  • The OFRR demonstrated quantitative detection of methylated DNA.
  • MBD-2 protein showed significantly higher binding affinity for dsDNA compared to ssDNA.
  • The anti-5-methylcytidine antibody exhibited stronger affinity for ssDNA than dsDNA.

Conclusions:

  • The OFRR is a promising platform for sensitive, label-free detection of DNA methylation.
  • MBD-2 protein is a suitable capture molecule for dsDNA methylation analysis using OFRR.
  • The choice of capture molecule (MBD-2 vs. antibody) depends on whether ssDNA or dsDNA methylation is being investigated.
