American Trypanosomiasis01:22

American Trypanosomiasis

Chagas disease, or American trypanosomiasis, is a vector-borne parasitic infection caused by Trypanosoma cruzi, a flagellated protozoan (kinetoplastid) of the family Trypanosomatidae. The disease is endemic in Latin America, although cases are increasingly reported worldwide due to human migration. Transmission most commonly occurs when feces of infected triatomine bugs contaminate bite wounds or mucosal surfaces; additional routes include congenital, transfusional, transplant-related, and oral...

Area of Science:

  • Parasitology
  • Cell Biology
  • Microscopy

Background:

  • Trypanosoma brucei causes African sleeping sickness in humans and cattle.
  • T. brucei is a model organism for kinetoplastid research due to ease of culture and genetic manipulation.

Purpose of the Study:

  • To describe sample preparation methods for T. brucei for transmission electron microscopy.
  • To introduce a novel high-pressure freezing protocol for improved sample quality.

Main Methods:

  • Conventional sample preparation: chemical fixation, detergent extraction, dehydration, and Epon embedding.
  • Novel method: high-pressure freezing followed by rapid freeze substitution and HM20 embedding.

Main Results:

  • Conventional methods provide a thorough explanation of T. brucei sample preparation.
  • High-pressure freezing yields samples with good cell morphology suitable for immunocytochemistry.

Conclusions:

  • Both conventional and high-pressure freezing methods are effective for preparing T. brucei for electron microscopy.
  • The high-pressure freezing protocol offers advantages for preserving cell morphology for immunocytochemical studies.
