Summary

Chronic myeloid leukemia (CML) has evolved from early 19th-century recognition to targeted therapies. Modern tyrosine kinase inhibitors (TKIs) like imatinib are now the preferred initial treatment for CML, replacing stem cell transplantation.

  • Chronic myeloid leukemia (CML) was first identified in the mid-19th century, initially treated with arsenic.
  • Early 20th-century treatments included radiotherapy, busulfan, hydroxycarbamide, and interferon-alfa (IFN-α).

Observation:

  • Key discoveries include the Philadelphia chromosome (1960), the (9;22) translocation (1973), the BCR breakpoint cluster region (1984), and the BCR-ABL transcript (1985).
  • Allogeneic stem cell transplantation (SCT) emerged as a primary treatment from 1980 onwards for eligible patients.

Findings:

  • The development of tyrosine kinase inhibitors (TKIs) starting in 1998 revolutionized CML treatment.
