Gastrulation01:56

Gastrulation

Gastrulation establishes the three primary tissues of an embryo: the ectoderm, mesoderm, and endoderm. This developmental process relies on a series of intricate cellular movements, which in humans transforms a flat, “bilaminar disc” composed of two cell sheets into a three-tiered structure. In the resulting embryo, the endoderm serves as the bottom layer, and stacked directly above it is the intermediate mesoderm, and then the uppermost ectoderm. Respectively, these tissue strata will form...

Summary

Oct1 (Pou2f1) is essential for early embryonic development, particularly for trophoblast stem cell maintenance and differentiation. Its loss leads to embryonic lethality before gastrulation, impacting the maternal-embryonic interface.

Area of Science:

  • Developmental Biology
  • Genetics
  • Molecular Biology

Background:

  • Oct1 (Pou2f1) is a ubiquitously expressed transcription factor.
  • Its known functions include regulating cellular responses to stress.
  • Its role in early embryonic development was previously unclear.

Purpose of the Study:

  • To investigate the essential function of Oct1 during early mouse embryonic development.
  • To characterize the phenotype of Oct1-null embryos.

Main Methods:

  • Generation and analysis of Oct1 loss-of-function mouse models.
  • Molecular and morphological analyses of mutant embryos.
  • Tetraploid complementation assay.

Main Results:

  • Oct1-null embryos exhibit early embryonic lethality at the streak stage.
  • Mutant embryos show defects in extra-embryonic ectoderm formation and ectoplacental cone development.
  • Oct1 is required for trophoblast stem cell maintenance and differentiation, with Cdx2 identified as a direct target.

Conclusions:

  • Oct1 is essential for post-implantation development, specifically for trophoblast stem cell function.
  • Oct1 plays a critical role in establishing the maternal-embryonic interface.
  • Oct1 is also required in the embryo proper from late gastrulation onwards.
