Background:

  • Gossypol, a compound derived from cotton seeds, has shown antifertility effects in male rats.
  • The mechanism and species-specific differences in gossypol's effects, particularly regarding its accumulation in organs, require further investigation.

Purpose of the Study:

  • To investigate the organ-specific accumulation of gossypol in male rabbits and rats.
  • To explore the correlation between gossypol accumulation in the testis and its antifertility effects, comparing rabbit and rat models.

Main Methods:

  • Male rabbits and rats were administered gossypol (20 mg/kg/day) for 12 and 7 weeks, respectively.
  • Gossypol levels in various organs were quantified using the Smith aniline method.

Main Results:

  • Spleen showed the highest gossypol accumulation in both species.
  • Rabbit testes accumulated significantly lower levels of gossypol compared to rat testes, despite longer administration in rabbits.
  • Gossypol levels in rabbit brain were minimal, and undetectable in rat brain, ruling out hypothalamic-pituitary axis involvement.

Conclusions:

  • Lower gossypol accumulation in rabbit testes may explain their non-sensitivity to its antifertility effects.
  • Gossypol's mechanism of action on the testis does not appear to involve the hypothalamus-pituitary axis due to negligible brain accumulation.
