Insights

Oral administration of β-1,3-glucan (Imunoglucan®) protected mice against lethal Listeria monocytogenes by boosting immune cell production and cytokine release, preventing infection-induced immune suppression.

Area of Science:

  • Immunology
  • Microbiology
  • Pharmacology

Background:

  • Listeria monocytogenes (LM) infection can cause severe immune suppression.
  • Oral administration of β-1,3-glucan has shown potential immunomodulatory effects.

Purpose of the Study:

  • To investigate the protective effects of oral β-1,3-glucan (Imunoglucan®) against Listeria monocytogenes infection in mice.
  • To elucidate the underlying mechanisms of Imunoglucan®'s immunomodulatory activity.

Main Methods:

  • Mice were prophylactically treated with varying doses of Imunoglucan® before lethal or sublethal LM challenge.
  • Survival rates, myelosuppression, splenomegaly, progenitor cell counts (CFU-GM), colony-stimulating activity (CSA), cytokine levels (IL-6, IL-1α, INF-γ), and stromal cell function were assessed.

Main Results:

  • Prophylactic Imunoglucan® (150 and 300 mg/kg) significantly increased survival rates (up to 40%) against lethal LM infection.
  • Effective doses prevented myelosuppression and splenomegaly by increasing bone marrow myeloid progenitors (CFU-GM) and serum colony-stimulating activity (CSA).
  • Imunoglucan® up-regulated IL-6, IL-1α, and INF-γ production and restored stromal cell function, counteracting LM-induced immune suppression.

Conclusions:

  • Oral Imunoglucan® confers protection against Listeria monocytogenes by indirectly modulating immune responses.
  • The mechanism involves enhancing myeloid progenitor reserves and promoting the release of key cytokines (CSFs, IL-1α, IL-6, INF-γ).
  • Imunoglucan® appears to counteract Listeria-induced immune suppression through these immunomodulatory effects.
