Summary

Researchers developed a simple method to produce monolysocardiolipin (MLCL) from cardiolipin (CL) using phospholipase A2. This process in methanol yields approximately 80% MLCL, with hydrolysis occurring specifically at the sn-2 position.

Area of Science:

  • Biochemistry
  • Lipidomics
  • Enzymology

Background:

  • Cardiolipin (CL) is a vital phospholipid in mitochondrial membranes.
  • Selective modification of CL is crucial for studying its functions.
  • Existing methods for producing lysocardiolipin derivatives can be inefficient.

Purpose of the Study:

  • To describe a simple, high-yielding preparation of monolysocardiolipin (MLCL).
  • To investigate the regioselectivity of cardiolipin hydrolysis by phospholipase A2 in methanol.
  • To provide characterized MLCL for further biochemical and biophysical studies.

Main Methods:

  • Enzymatic hydrolysis of cardiolipin (CL) using phospholipase A2 in methanol.
  • Semi-preparative scale synthesis of monolysocardiolipin (MLCL).

  • Characterization of synthesized lipids using Nuclear Magnetic Resonance (NMR) and Electrospray Ionization-Mass Spectrometry/Mass Spectrometry (ESI-MS/MS).
  • Main Results:

    • Phospholipase A2 selectively hydrolyzes cardiolipin in methanol, preferentially yielding monolysocardiolipin.
    • The reaction achieved an approximate 80% yield of monolysocardiolipin.
    • Hydrolysis was confirmed to occur exclusively at the sn-2 position of cardiolipin.

    Conclusions:

    • A straightforward and efficient method for preparing monolysocardiolipin from cardiolipin is established.
    • The regioselective hydrolysis of cardiolipin by phospholipase A2 in methanol offers a valuable synthetic route.
    • The characterized monolysocardiolipin is suitable for diverse lipid research applications.
