Pathogen reduction technology (PRT) effectively preserves platelet concentrate (PC) properties in additive solutions for 5 days. PASIIIM better maintains platelet function than PASIII for 7-day storage.
Area of Science:
- Hematology
- Transfusion Medicine
- Biotechnology
Background:
- Limited research exists on pathogen reduction technology (PRT) effects on platelet (PLT) products stored in platelet additive solutions (PASs).
- This study evaluates Mirasol PRT and storage in PAS (PAS-III or PAS-IIIM) on platelet concentrate (PC) characteristics.
Purpose of the Study:
- To assess the impact of Mirasol PRT on analytical and functional parameters of platelet concentrates stored in PAS.
- To compare the efficacy of PAS-III and PAS-IIIM in preserving PRT-treated platelet concentrates.
Main Methods:
- Apheresis-derived PCs were divided into untreated (plasma or PAS storage) and PRT-treated (PAS storage) groups.
- Evaluated cell quality (pH, swirl, lactate, glucose), flow cytometry, and perfusion studies over 7 days.
- Assessed adhesive and aggregating functions using a perfusion system.
Main Results:
- Cell quality variables declined during storage; swirl was better maintained in plasma.
- PRT-treated PCs showed moderate glycoprotein Ib reduction by Day 7.
- PRT-treated PCs in PAS demonstrated comparable subendothelial reactivity to untreated PCs at Day 5.
- PASIIIM better preserved PRT-PC cohesive properties at Day 7 compared to PASIII.
Conclusions:
- Platelet properties are maintained in PRT-treated concentrates stored in PAS for up to 5 days.
- PASIIIM offers superior preservation of adhesive and cohesive functions for PRT-PCs over 7 days compared to PASIII.
- Further clinical studies are warranted to validate these in vitro findings.

Formation of the Platelet Plug
As the injured blood vessel contracts, endothelial cells undergo contraction, revealing collagen fibers in the basement membrane and underlying connective tissue. Furthermore, the plasma membrane of endothelial cells becomes adhesive, preparing the site for platelet adhesion. Platelets...

Structure and Function of Platelets
Platelets are continually replenished, circulating in the bloodstream for 9-12 days before being removed by phagocytes, primarily in the spleen. A microliter of circulating blood contains between 150,000 and 450,000 platelets, with...
