Purpose of the Study:

  • To investigate the pharmacokinetic profile of 4-hydroxyanisole (4HA) in patients with lower limb malignant melanoma.
  • To assess changes in 4HA pharmacokinetics with repeated administration.

Main Methods:

  • Ten grams of 4HA was intra-arterially infused twice daily into ten female patients.
  • Serum 4HA concentrations were measured after the first and fourth infusions.
  • Pharmacokinetic parameters including half-life (t1/2), area under the curve (AUC), volume of distribution (VD), and clearance were analyzed.

  • Serum 4HA concentrations declined biphasically with distribution and elimination half-lives of 6.3 and 70.9 min, respectively.

Conclusions:

  • Short-term enzyme induction does not appear to influence 4-hydroxyanisole clearance.
  • Changes in the volume of distribution over time necessitate potential adjustments in 4HA dosage regimens for melanoma patients.
