Summary

Healthy human blood monocytes do not contain pre-made tumor necrosis factor (TNF)-alpha. Staphylococcus aureus can induce TNF-alpha mRNA, but this process is blocked by cycloheximide.

Area of Science:

  • Immunology
  • Molecular Biology
  • Cell Biology

Background:

  • Tumor necrosis factor-alpha (TNF-alpha) is a key inflammatory cytokine.
  • Understanding TNF-alpha regulation in human monocytes is crucial for immune response studies.

Purpose of the Study:

  • To determine the presence of TNF-alpha mRNA and protein in circulating human blood monocytes.
  • To investigate TNF-alpha gene expression in isolated human monocytes.
  • To analyze the effect of cycloheximide on TNF-alpha induction.

Main Methods:

  • Isolation of human monocytes using continuous Percoll gradient fractionation.
  • Direct RNA isolation from blood samples for mRNA expression analysis.
  • Detection of TNF-alpha mRNA and protein.

Related Experiment Videos

Main Results:

  • Human blood leukocytes from healthy donors lack pre-existing TNF-alpha mRNA and protein.
  • Staphylococcus aureus can induce TNF-alpha mRNA expression.
  • Cycloheximide pretreatment inhibits Staphylococcus aureus-induced TNF-alpha mRNA.

Conclusions:

  • Circulating human monocytes do not constitutively express TNF-alpha.
  • TNF-alpha gene expression is inducible in human monocytes.
  • Protein synthesis inhibition affects TNF-alpha mRNA induction pathways.
