Gene interactions influence attention performance in children with attention-deficit/hyperactivity disorder. Specifically, the adrenergic α-2A receptor (ADRA2A) and brain-derived neurotrophic factor (BDNF) genes interact, affecting response time variability on the continuous performance test (CPT).
Area of Science:
- Neuroscience
- Genetics
- Psychiatry
Background:
- Complex phenotypes like continuous performance test (CPT) performance often involve gene-gene interactions (epistasis).
- Genetic variations in the noradrenergic system and brain-derived neurotrophic factor (BDNF) have been linked to CPT performance.
- BDNF plays a crucial role in the development and survival of noradrenergic neurons.
Purpose of the Study:
- To investigate the gene-gene interaction between the adrenergic α-2A receptor (ADRA2A) and BDNF on CPT performance.
- To examine this interaction in a Korean pediatric population diagnosed with attention-deficit/hyperactivity disorder (ADHD).
Main Methods:
- Genotyping of ADRA2A (rs583668) and BDNF (rs11030101) polymorphisms in 122 Korean children with ADHD.
- Assessment of CPT performance, including response time variability.
- Statistical analysis to determine the interaction effect between ADRA2A and BDNF polymorphisms.
Main Results:
- A significant gene-gene interaction was identified between ADRA2A rs553668 and BDNF rs11030101.
- This interaction significantly affected response time variability during the CPT (P=0.011).
Conclusions:
- The study provides preliminary evidence for a significant interaction between ADRA2A and BDNF genes influencing CPT performance in children with ADHD.
- This finding highlights the potential role of combined genetic factors in the neurobiological underpinnings of ADHD-related cognitive deficits.

Background and Environment Affect Phenotype
An example of how genetic background affects phenotype can be seen in horses. The Extension gene in horses is responsible for their coat color. A wild-type gene (EE) produces black pigment in the coat, while a mutant gene (ee) produces red pigment. A...
