Area of Science:

  • Virology
  • Structural Biology
  • Microscopy

Background:

  • Hantaan virus is a prototypic member of the Hantavirus genus (family Bunyaviridae), causing hemorrhagic fever with renal syndrome.
  • Bunyaviridae are negative-sense RNA viruses with segmented genomes, enveloped virions, and glycoprotein spikes (Gn and Gc).

Purpose of the Study:

  • To determine the structural organization of Hantaan virus virions using advanced microscopy techniques.
  • To elucidate the arrangement of surface glycoproteins and internal components within the virion.

Main Methods:

  • Cryo-electron tomography (cryo-ET) and single-particle cryo-electron microscopy (cryo-EM).
  • High-resolution structural determination of the Gn-Gc spike complex.

Main Results:

  • The tetrameric Gn-Gc spike complex structure was resolved to 2.5 nm, showing ordered lattices on the virion surface.
  • Cytoplasmic extensions of Gn-Gc spikes also formed a lattice on the inner viral membrane.
  • Internal rod-shaped ribonucleoprotein complexes were arranged in parallel pairs and triplets, differing from icosahedral structures in some bunyaviruses.

Conclusions:

  • Hantaan virus exhibits a distinct structural organization compared to some other bunyaviruses.
  • The findings suggest a common structural framework among hantaviruses, as indicated by comparison with Tula hantavirus.
  • This structural information is crucial for understanding hantavirus biology and pathogenesis.
