Sample Preparation for Analysis: Overview01:21

Sample Preparation for Analysis: Overview

Sample preparation is an essential step in the analytical process. It involves preparing a sample so that it can be analyzed accurately. The goal is to extract the analyte, the substance you want to measure, from the sample while removing any components that may interfere with the analysis. Sample preparation techniques vary depending on the physical state of the sample.
Bulk or large solid samples are typically reduced in size using grinding, crushing, or milling techniques to increase the...

Summary

Developing a liquid chromatography-tandem mass spectrometry (LC-MS/MS) assay using dried blood spot (DBS) samples requires optimized sample preparation. Liquid-liquid extraction demonstrated the least ion suppression/enhancement for DBS LC-MS/MS analysis.

Area of Science:

  • Analytical Chemistry
  • Biomarker Analysis

Background:

  • Dried blood spot (DBS) sampling is increasingly adopted for bioanalysis.
  • Standardized methods for developing and validating LC-MS/MS assays with DBS are needed.

Purpose of the Study:

  • To evaluate three distinct DBS sample preparation techniques for LC-MS/MS assay development.
  • To introduce and assess the term 'elution efficiency' in DBS analysis.

Main Methods:

  • Comparison of organic extraction, protein precipitation, and liquid-liquid extraction for DBS sample preparation.
  • Evaluation of three different DBS card types.
  • Development, qualification, and application of a DBS LC-MS/MS method.

Main Results:

  • Liquid-liquid extraction yielded the least ion suppression/enhancement.
  • Protein precipitation and liquid-liquid extraction effectively eluted the probe compound.
  • Organic extraction using pure solvents showed low elution efficiency.
  • Significant ion suppression/enhancement was observed with organic extraction and protein precipitation on specific card types.

Conclusions:

  • Liquid-liquid extraction is the preferred method to minimize ion suppression/enhancement in DBS LC-MS/MS assays.
  • Elution efficiency is a critical parameter for assessing DBS sample preparation effectiveness.
  • The choice of DBS card and sample preparation method significantly impacts assay performance.
