Insulin: The Receptor and Signaling Pathways01:28

Insulin: The Receptor and Signaling Pathways

Insulin action is mediated through a receptor tyrosine kinase, akin to the IGF-1 receptor. The number of receptors per cell varies significantly, from 40 on erythrocytes to 300,000 on adipocytes and hepatocytes. The insulin receptor consists of linked α/β subunit dimers, forming a heterotetramer glycoprotein with two extracellular α subunits and two β subunits spanning the membrane. The α subunits inhibit the inherent tyrosine kinase activity of the β subunits, but this inhibition is released...

Transducer Mechanism: Enzyme-Linked Receptors

Enzyme-linked receptors are cell-surface receptors acting as an enzyme or associating with an enzyme intracellularly. They make excellent drug targets. Drugs can bind to the extracellular ligand-binding domain or directly affect their enzymatic domain and alter their activity.
Major types that are helpful drug targets include:
Intracellular Hormone Receptors01:08

Intracellular Hormone Receptors

Lipid-soluble hormones diffuse across the plasma and nuclear membrane of target cells to bind to their specific intracellular receptors. These receptors act as transcription factors that regulate gene expression and protein synthesis in the target cell
Spare Receptors01:30

Spare Receptors

Some receptors remain unoccupied even when an agonist produces a maximal response. Such empty ones are called spare receptors. In presence of spare receptors the maximum effect of an agonist drug is achieved with fewer than 100% of the receptors being occupied. To determine the presence of spare receptors, scientists often compare the concentration of the drug needed to produce 50% of the maximum effect (EC50) with the concentration of the drug needed to occupy 50% of the receptors (Kd). If the...

Overview
Receptor-mediated Endocytosis01:20

Receptor-mediated Endocytosis

Receptor-mediated endocytosis is when bulk amounts of specific molecules are imported into a cell after binding to cell surface receptors. The molecules bound to these receptors are taken into the cell through inward folding of the cell surface membrane, which is eventually pinched off into a vesicle within the cell. Structural proteins, such as clathrin, coat the budding vesicle.
Clathrin-Mediated Endocytosis of LDL
One well-characterized example of receptor-mediated endocytosis is the...

Summary

Leptin receptor (Ob-R) has multiple isoforms, with the long Ob-Rb isoform primarily regulating energy balance and immunity. Short isoforms bind Janus kinases, but only Ob-Rb fully signals through the JAK/STAT pathway.

Area of Science:

  • Endocrinology
  • Immunology
  • Molecular Biology

Background:

  • The leptin receptor (Ob-R) is a class I cytokine receptor crucial for regulating energy homeostasis.
  • Ob-R is encoded by the db gene and exists in six isoforms generated by alternative RNA splicing.
  • Isoforms are classified as long, short, or secretory, differing in structure and function.

Purpose of the Study:

  • To elucidate the distinct roles and signaling capabilities of various leptin receptor (Ob-R) isoforms.
  • To understand how different Ob-R isoforms contribute to leptin's functions in metabolism and immunity.

Main Methods:

  • Analysis of Ob-R gene structure and alternative splicing.
  • Characterization of structural differences among Ob-R isoforms.
  • Investigation of Ob-R isoform interactions with signaling molecules like JAK kinases.

Main Results:

  • The long Ob-Rb isoform is primarily expressed in the hypothalamus, regulating energy balance and secretory organs.
  • Ob-Rb is also expressed on immune cells, influencing innate and adaptive immunity.
  • Short isoforms (Ob-Ra, Ob-Rc, Ob-Rd) possess the box 1 motif for JAK kinase binding and signal transduction.
  • A soluble isoform (Ob-Re) may modulate serum leptin levels and act as a carrier protein.
  • The JAK/STAT pathway is central to leptin signaling via membrane receptors.

Conclusions:

  • Only the full-length Ob-Rb isoform is capable of complete signal transduction.
  • Different Ob-R isoforms exhibit specialized functions in metabolic and immune processes.
  • Understanding Ob-R isoform diversity is key to comprehending leptin's multifaceted biological roles.
