Background:

  • Abscisic acid (ABA) regulates crucial plant processes like seed germination and stress responses.
  • ABA-mediated gene expression involves ABA response elements (ABREs) and coupling elements (CEs).
  • While ABRE-binding factors are known, the proteins interacting with CEs remained unidentified.

Purpose of the Study:

  • To identify transcription factors that bind to the CE1 coupling element.
  • To investigate the in vivo functions of CE1-binding factors (CEBFs) in plant responses.

Main Methods:

  • Yeast one-hybrid screens were employed to isolate CE1-binding proteins from ABA-treated Arabidopsis seedlings.
  • Reporter genes with a CE1 trimer were used in yeast transformation screens.
  • Analysis of positive clones identified AP2/ERF domain proteins as CE1 binders.

Main Results:

  • A group of AP2/ERF domain proteins were identified as binding to the CE1 element.
  • One CEBF, AtERF13, was found to confer ABA hypersensitivity in Arabidopsis.
  • Two other CEBFs were shown to enhance sugar sensitivity in overexpression studies.

Conclusions:

  • AP2/ERF superfamily proteins interact with the CE1 coupling element.
  • CEBFs play roles in abscisic acid and/or sugar responses.
  • CE1 may function as both an ABRE and an element involved in biotic/abiotic stress responses.
