T Cell Types and Functions

When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. The differentiation of CD4 T cells into helper T cell subsets, such as Th1, Th2, and Th17 cells, is dependent on the antigen type, antigen-presenting cell, and regulatory cytokines.

Background:

  • This study examined thymic cellular output in hypogonadal men receiving androgen replacement therapy.

Purpose of the Study:

  • To investigate the relationship between hypogonadism and thymic T-cell output.
  • To assess the effects of androgen replacement therapy on thymic function.

Main Methods:

  • Quantified circulating naive T cells (CD4+CD45RA+) via flow cytometry.
  • Measured T-cell receptor excision circles using real-time PCR in peripheral blood mononuclear cells.
  • Studied healthy men and hypogonadal men before and after testosterone replacement.

Main Results:

  • Hypogonadal men showed significantly increased proportions of naive T cells (35.5% and 44.4%) compared to healthy males (10.5%).
  • Testosterone therapy in one patient reduced naive T cells from 37.36% to 12.51% over 7 months.
  • T-cell receptor excision circle levels decreased by 78% and 83% after androgen replacement in two patients.

Conclusions:

  • The hypogonadal state is linked to elevated thymic T-cell output.
  • Androgen replacement therapy effectively reverses the increased thymic output of recent thymic emigrants in hypogonadal men.
