Context:

  • Nuclear factor-kappa B (NF-kB) is a transcription factor implicated in cellular resistance to tumor necrosis factor-alpha (TNF-α)-induced apoptosis.
  • Prostate-specific antigen (PSA) is a key biomarker for prostate health and disease.

Purpose:

  • To investigate the relationship between the expression of NF-kB subunits (p50 and p65) and serum PSA levels.
  • To compare NF-kB expression in normal prostate tissue, benign prostatic hyperplasia (BPH), and prostate cancer (PC).

Summary:

  • NF-kB p50 and p65 subunits were evaluated in 5 normal, 24 BPH, and 19 PC patient samples using immunohistochemical and Western blot analyses.
  • While p65 was absent and p50 showed limited cytoplasmic expression in controls, both subunits were detected in 62.5% of BPH and 63.2% of PC samples.
  • Increased expression frequency of both p50 and p65 correlated with higher serum PSA levels across all groups.

Impact:

  • Findings suggest that NF-kB activation, indicated by nuclear translocation in prostate cancer, may be linked to disease progression and elevated PSA.
  • Understanding the biological mechanisms connecting circulating PSA levels and NF-kB expression is crucial.
