Small supernumerary marker chromosomes (sSMC) can cause abnormal phenotypes, but are hard to identify. Advanced molecular techniques accurately characterize sSMC, aiding genotype-phenotype correlation in patients.
Area of Science:
- Genetics
- Molecular Biology
- Human Genetics
Background:
- Small supernumerary marker chromosomes (sSMC) are additional, structurally altered chromosomes.
- Conventional karyotyping may not fully identify sSMC, complicating clinical correlation, especially prenatally.
- Approximately one-third of sSMC carriers exhibit abnormal phenotypes.
Purpose of the Study:
- To characterize 19 sSMC in 15 patients with dysmorphic features and/or congenital anomalies.
- To evaluate the utility of advanced molecular cytogenetic techniques for sSMC characterization.
- To improve genotype-phenotype correlation and risk assessment for sSMC cases.
Main Methods:
- Routine cytogenetics with banding techniques for initial sSMC identification.
- Advanced characterization using array comparative genomic hybridization (array-CGH).
- Fluorescence in situ hybridization (FISH) techniques including multicolor banding (MCB), cenM-FISH, subcenM-FISH, micro-dissection, and reverse FISH.
Main Results:
- Successfully characterized 19 sSMC using a combination of conventional and advanced molecular methods.
- Demonstrated the effectiveness of array-CGH and various FISH techniques in detailed sSMC analysis.
- Identified specific sSMC in patients presenting with dysmorphic features, congenital anomalies, short stature, or ambiguous genitalia.
Conclusions:
- Advanced molecular cytogenetic techniques are crucial for accurate sSMC characterization.
- These methods enhance the ability to correlate sSMC genotype with patient phenotype.
- Improved characterization aids in understanding the clinical presentation and risks associated with sSMC.

Proteomics
Proteomics is the study of proteomes' function. It involves the large-scale systematic study of the proteome to denote the protein complement expressed by a genome. Scientist Mark Wilkins coined the term proteomics...
