Interleukin-4 (IL-4) gene promoter polymorphisms may increase Behçet
Area of Science:
- Immunogenetics
- Rheumatology
- Molecular Biology
Background:
- Cytokine gene polymorphisms influence individual susceptibility to diseases like Behçet's Disease (BD).
- Interleukin-4 (IL-4) and its receptor alpha (IL-4Rα) are key immune modulators.
- Understanding genetic associations can elucidate BD pathogenesis.
Purpose of the Study:
- To investigate the association between IL-4 and IL-4Rα gene polymorphisms and susceptibility to Behçet's Disease (BD) in a Turkish population.
- To explore the potential role of these polymorphisms in disease risk and clinical manifestations, such as Pathergy test positivity.
Main Methods:
- Genotyping of IL-4 and IL-4Rα gene polymorphisms was performed using Polymerase Chain Reaction (PCR) sequence-specific primers.
- DNA samples were analyzed from 97 BD patients and 76 healthy controls from a Turkish cohort.
- Allele and genotype frequencies were compared between patient and control groups.
Main Results:
- Higher frequencies of IL-4 gene promoter genotypes (-1098 TG and -590 CT) and alleles (-1098 G and -590 T) were observed in BD patients compared to controls.
- Specific IL-4 haplotypes (TTC) were found to be associated with BD.
- IL-4Rα gene polymorphism showed a potential association with Pathergy test positivity in BD patients.
Conclusions:
- IL-4 gene promoter polymorphisms may contribute to susceptibility to Behçet's Disease (BD).
- These genetic variations might increase the risk of developing BD.
- Further large-scale studies are required to confirm the association of IL-4 and IL-4Rα polymorphisms with BD.
