T Cell Types and Functions

When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. The differentiation of CD4 T cells into helper T cell subsets, such as Th1, Th2, and Th17 cells, is dependent on the antigen type, antigen-presenting cell, and regulatory cytokines.

Background:

  • Interleukin-27 (IL-27) and Interleukin-23 (IL-23) are critical heterodimeric cytokines involved in immune responses and disorders.
  • While IL-27 is known to affect mature B cells, its specific role in human plasma cells (PC) and plasmablasts (PPC) is not well-established.
  • The function of IL-23 in human PC and PPC also requires further investigation.

Purpose of the Study:

  • To investigate the functional roles of IL-27 and IL-23 on human PC and PPC.
  • To determine if these cytokines modulate key functions such as cell migration and immunoglobulin production in human PC and PPC.

Main Methods:

  • Utilized in vitro generated human PPC, and PC isolated from human tonsils and bone marrow (BM).
  • Assessed the expression of IL-27 receptor (IL-27R) and IL-23 receptor (IL-23R) on PC and PPC.
  • Evaluated the effects of IL-27 and IL-23 on chemokine/chemokine receptor modulation, immunoglobulin (IgM, IgG) secretion, and cell chemotaxis.

Main Results:

  • Human PC and PPC express the complete IL-27 receptor (IL-27R).
  • IL-27 demonstrated chemotactic properties on PC and PPC, modulating chemokines and chemokine receptors, and influencing IgM and IgG secretion.
  • Human PC and PPC express both chains of IL-23 receptor (IL-23R) and IL-23; IL-23 similarly affected Ig production but did not induce PC chemotaxis.

Conclusions:

  • This study provides the first evidence that IL-27 and IL-23 act on human PC and PPC.
  • IL-27 and IL-23 play significant roles in modulating immunoglobulin production by human PC and PPC.
  • The findings offer novel insights into the involvement of IL-27 and IL-23 in human B cell immune responses.
