Purpose of the Study:
- To investigate the potential role of B19 nonstructural protein 1 (NS1) in the development of autoimmune thyroiditis.
- To examine if the B19 NS1 gene can induce thyroiditis in a nonpermissive genetic background.
Main Methods:
- Transgenic C57BL/6 mice expressing the B19 NS1 gene were developed.
- Mice were immunized with thyroglobulin (Tg) or saline.
- Thyroiditis incidence, intrathyroidal T-cell receptor gene expression, and anti-Tg antibody levels were assessed.
Main Results:
- Thyroiditis was not observed in saline-treated mice or Tg-immunized males.
- Tg immunization induced thyroiditis in 25% of transgenic females, but not wild-type females.
- No significant differences in thyroiditis incidence, gene expression, or antibody levels were found between transgenic and wild-type females.
Conclusions:
- The B19 NS1 gene alone is insufficient to induce autoimmune thyroiditis in this mouse model.
- Further studies are necessary to definitively exclude the involvement of B19 infection in HT pathogenesis.
