The Early Endosome: Endocytosis of Transferrin01:28

The Early Endosome: Endocytosis of Transferrin

Essential proteins such as insulin or low-density lipoprotein (LDL) and micronutrients such as iron enter a eukaryotic cell through receptor-mediated endocytosis. Subsequently, the early endosomes fuse with the vesicles containing such receptor-ligand complexes and play a vital role in sorting the incoming ligands and receptors. While the ligands are either degraded inside the vesicle or released into the cytosol, their receptors are returned to the plasma membrane for further rounds of...

Summary

Pathogenic bacteria like Neisseria meningitidis use iron-acquisition systems to obtain iron from transferrin (Tf). Tf-binding proteins A and B (TbpA and TbpB) act as the cell-surface Tf receptor in these bacteria.

Area of Science:

  • Microbiology
  • Pathogen-host interactions
  • Molecular biology

Background:

  • Pathogenic bacteria require iron, a vital nutrient, for survival and virulence.
  • Iron acquisition systems are crucial for pathogens to obtain iron during infection.
  • Transferrin (Tf) is the primary glycoprotein for extracellular iron transport in vertebrates.

Purpose of the Study:

  • To investigate the role of iron-acquisition systems in pathogens.
  • To identify the function of Tf-binding proteins A and B (TbpA and TbpB) in Neisseria meningitidis.

Main Methods:

  • Analysis of bacterial iron-acquisition systems.
  • Identification of transferrin (Tf) as a target for iron uptake.
  • Characterization of TbpA and TbpB as cell-surface Tf receptors.

Main Results:

  • Neisseria meningitidis utilizes iron-acquisition systems to scavenge iron.
  • Tf-binding proteins A and B (TbpA and TbpB) are cell-surface receptors for transferrin (Tf).

Conclusions:

  • TbpA and TbpB are essential components of the Neisseria meningitidis iron-acquisition system.
  • Understanding these receptors can provide insights into pathogen virulence mechanisms.
