This study found no significant link between hypothalamic gene variations and weight change. However, a specific gene variant (NMB SNP rs7180849) showed interaction with high glycemic index diets, suggesting a potential influence on weight gain.
Area of Science:
- Genetics and Nutrigenomics
- Obesity Research
- Metabolic Pathways
Background:
- Single nucleotide polymorphisms (SNPs) in hypothalamic pathway genes may influence weight gain.
- Dietary factors might modify the effects of these genetic predispositions.
Purpose of the Study:
- Investigate associations between SNPs in candidate genes and weight change over approximately 7 years.
- Examine potential effect modification by dietary glycemic index (GI) and protein intake.
Main Methods:
- A case-cohort study design was employed with over 50,000 participants from five European countries.
- 134 SNPs across 15 candidate genes were genotyped and analyzed for association with weight gain.
- Logistic and linear regression models were used to assess SNP associations and dietary interactions.
Main Results:
- No significant associations were found between the studied SNPs in hypothalamic genes and overall weight change.
- No significant effect modification by dietary factors was observed, except for one SNP (rs7180849 in NMB).
- Carriers of the minor allele for NMB SNP rs7180849 exhibited greater weight gain with higher dietary GI (P=2x10⁻⁷).
Conclusions:
- The study found no evidence linking SNPs in the investigated hypothalamic genes to significant weight change.
- The identified interaction between NMB SNP rs7180849 and dietary GI warrants further investigation and confirmation.
