Imaging Biological Samples with Optical Microscopy01:18

Imaging Biological Samples with Optical Microscopy

Optical microscopy uses optic principles to provide detailed images of samples. Antonie van Leeuwenhoek designed the first compound optical microscope in the 17th century to visualize blood cells, bacteria, and yeast cells. In 1830, Joseph Jackson Lister created an essentially modern light microscope. The 20th century saw the development of microscopes with enhanced magnification and resolution.
In optical microscopy, the specimen to be viewed is placed on a glass slide and clipped on the stage...

Area of Science:

  • Biomedical Engineering
  • Computational Pathology
  • Digital Pathology

Background:

  • Accurate cell counting is crucial for diagnosing neoplasms.
  • Traditional methods for counting cellular structures like AgNORs are time-consuming and subjective.
  • Computer-assisted image analysis offers potential for objective and efficient quantification.

Purpose of the Study:

  • To develop and evaluate image processing software for computer-assisted cellular structure counting.
  • To apply the software for quantitative analysis of AgNORs in ameloblastomas.
  • To compare the software's performance against manual counting by experienced pathologists.

Main Methods:

  • Development of image processing software with analytical tools for cell counting.
  • Codification of expert pathologist knowledge into automated processing steps.
  • Quantitative analysis of AgNORs per cell in ameloblastoma samples.
  • Comparison of automated results with manual visual analysis by calibrated observers.

Main Results:

  • The software successfully automated the estimation of mean AgNORs per cell.
  • No statistically significant difference (p<0.05) was found between computer-assisted and visual analysis.
  • The automated method provided accurate and precise data.

Conclusions:

  • The developed software is a valuable tool for estimating cell and cellular structure counts.
  • It offers an objective, efficient, and accurate alternative to traditional manual counting methods.
  • The software can aid in the study of both malignant and benign neoplasms.
