Summary

This meta-analysis found no significant link between Tumor Necrosis Factor-alpha (TNF-α) gene polymorphisms and overall breast cancer risk. However, a negative association was observed between TNF-α-308 polymorphism and breast cancer in Caucasian populations.

Synthesis and Characterization of an Aspirin-fumarate Prodrug that Inhibits NFκB Activity and Breast Cancer Stem Cells

Background:

  • Tumor Necrosis Factor-alpha (TNF-α) is a pro-inflammatory cytokine implicated in cancer development.
  • TNF-α gene polymorphisms are known to affect cancer risk, but their association with breast cancer (BC) is debated.

Purpose of the Study:

  • To conduct a meta-analysis for precise estimation of the association between TNF-α polymorphisms (-238, -308, -857, -863, -1031, -1210) and breast cancer risk.

Main Methods:

  • Systematic electronic database searches were performed for relevant studies published up to March 2011.
  • Meta-analysis utilized Odds Ratios (OR) with 95% Confidence Intervals (CI) in fixed- and random-effect models.
  • Included 17 studies encompassing 44,442 BC patients and 49,926 controls.

Main Results:

  • No significant association was found between TNF-α-308 polymorphism and overall or Asian BC risk.
  • A negative association was observed between TNF-α-308 polymorphism and BC risk in Caucasian populations.
  • No significant associations were detected for TNF-α-238, -857, -863, -1031, or -1210 polymorphisms with BC risk in any population subgroup.

Conclusions:

  • The studied TNF-α gene polymorphisms (-308, -238, -857, -863, -1031, -1210) are not significantly associated with overall breast cancer risk.
  • A potential negative association exists between TNF-α-308 polymorphism and breast cancer risk specifically within Caucasian populations.
