Genetic links to infant temperament were explored using neonatal behavior and gene polymorphisms. The SLC6A4, DRD4, and TFAP2B genes showed associations, including gene-gene interactions, influencing infant behavioral traits.
Area of Science:
- Behavioral Genetics
- Neuroscience
- Molecular Biology
Background:
- Human personality genetics are complex, yielding limited results.
- Neonatal temperament offers an alternative phenotype for studying human behavior genetics.
- Observational measures of infant temperament can provide valuable insights.
Purpose of the Study:
- To investigate the genetic underpinnings of neonatal temperament.
- To identify specific gene polymorphisms associated with behavioral traits in newborns.
- To explore potential gene-gene interactions influencing temperament.
Main Methods:
- An association study on 117 Caucasian newborns using the Neonatal Behavior Assessment Scale.
- Genotyping of 13 polymorphisms in SLC6A4, DRD4, and TFAP2B genes.
- Reporter gene assays and quantitative PCR to assess functional effects of TFAP2B polymorphism.
Main Results:
- Significant associations found between SLC6A4 (5-HTTLPR, rs25531) and TFAP2B polymorphisms with the Range of State cluster.
- A gene-gene interaction between rs25531 and TFAP2B Indel Intron 2 was observed.
- DRD4 variable number tandem repeat exon 3 associated with orientation; TFAP2B polymorphism affected gene transcription in vitro.
Conclusions:
- The SLC6A4, DRD4, and TFAP2B genes play a role in neonatal temperament.
- Evidence supports a gene-gene interaction between SLC6A4 and TFAP2B.
- A TFAP2B polymorphism influences TFAP2B gene transcription, impacting behavior.
