Patch Clamp01:18

Patch Clamp

Many fundamental cell functions such as muscle contraction and nerve transmission rely on the electrical signals produced by the movement of positively and negatively charged ions across the cell membrane. One competent method to record current flowing across the whole cell or single ion channel is the patch-clamp technique.
In this method, a glass micropipette containing electrolyte solution is tightly sealed against a small portion of the cell membrane. As a result, a patch of the cell...

Area of Science:

  • Dermatology
  • Allergology
  • Immunology

Background:

  • Patch testing is the primary diagnostic tool for allergic contact dermatitis.
  • Standardization efforts have historically aimed to improve patch test validity and reliability.

Purpose of the Study:

  • To review modifications of the basic patch test technique for enhanced sensitivity.
  • To introduce and evaluate a standardized protocol for the strip patch test.

Main Methods:

  • Review of patch test modifications.
  • Development and testing of a standardized strip patch test protocol.
  • Comparison of strip patch test sensitivity and specificity against the traditional patch test.

Main Results:

  • The standardized strip patch test demonstrated increased test sensitivity compared to the standard patch test.
  • The protocol resulted in uniform stratum corneum reduction and good inter-rater agreement.
  • Evidence of non-specific inflammatory epidermal stimulation was observed.

Conclusions:

  • The standardized strip patch test significantly improves diagnostic accuracy as a complementary measure to patch testing.
  • This method shows vastly better sensitivity with only marginally lower specificity than the patch test.
  • Further research is required to confirm the reproducibility of the standardized strip patch test.
