Area of Science:

  • Reproductive Biology
  • Molecular Endocrinology
  • Oncogenesis

Background:

  • Sloan-Kettering virus gene product (Ski) is a nuclear proto-oncoprotein involved in cell differentiation and transformation.
  • Ski/SnoN proteins regulate the transforming growth factor-β (TGF-β) superfamily, key in follicle development.
  • SnoN's importance in follicle development suggests a role for Ski.

Purpose of the Study:

  • To investigate the expression patterns of Ski during mouse ovarian follicle development.
  • To clarify the relationship between Ski expression and key reproductive processes like ovulation and luteinization.

Main Methods:

  • Immunohistochemistry was used to detect Ski protein localization in ovarian tissues.
  • Real-time PCR quantified Ski mRNA levels in relation to specific reproductive stages.

Main Results:

  • Ski localized to oocyte nuclei in early development, shifting to the perinuclear space as follicles matured.
  • Ski expression varied across the estrous cycle and after gonadotropin stimulation, appearing in the corpus luteum post-luteinization.
  • Ski mRNA levels inversely correlated with ovulation-related genes during cumulus expansion and follicle development.

Conclusions:

  • Ski exhibits specific expression patterns throughout mouse ovarian follicle development, ovulation, and luteinization.
  • Ski likely plays critical roles in these reproductive processes, particularly during early follicular development.
