Summary

Growth hormone (GH) administration mitigates muscle damage during limb lengthening procedures. GH treatment enhanced muscle regeneration and reduced fibrogenesis in lengthened limbs, preserving muscle function.

Background:

  • Limb lengthening procedures can cause significant muscle damage and functional impairment.
  • Growth hormone (GH) plays a crucial role in tissue growth and repair, but its effects on muscle during limb lengthening are not fully understood.

Purpose of the Study:

  • To investigate the impact of growth hormone (GH) on muscle tissue during tibial lengthening in a rat model.
  • To assess GH's potential to reduce muscle damage and enhance muscle regeneration.

Main Methods:

  • Tibial lengthening was performed on 60 rats, with 30 receiving recombinant GH and 30 receiving saline.
  • Histopathological, immunohistochemical (vimentin, proliferating cell nuclear antigen, desmin), and electrophysiological analyses were conducted on the tibialis anterior muscle.

Main Results:

  • GH-treated lengthened limbs showed reduced muscle injury and lower vimentin expression compared to saline-treated limbs.
  • Cell proliferation (PCNA staining) and desmin expression were highest in GH-treated lengthened limbs, indicating enhanced myogenesis.
  • Electrophysiological studies demonstrated better muscle function retention in GH-treated lengthened limbs.

Conclusions:

  • Growth hormone (GH) effectively reduces muscle damage associated with limb lengthening.
  • GH promotes myogenesis and decreases fibrogenesis, thereby improving muscle outcomes during limb lengthening procedures.
