Summary

Versican, a key proteoglycan in the trabecular meshwork (TM), influences aqueous humor outflow. Its silencing altered outflow facility differently in human and porcine eyes, suggesting species-specific roles in regulating eye pressure.

  • Versican is a large proteoglycan with chondroitin sulfate (CS) glycosaminoglycan (GAG) side chains.

Purpose of the Study:

  • To investigate versican's distribution in the trabecular meshwork (TM).
  • To determine versican's effect on aqueous humor outflow facility.

Main Methods:

  • Used fluorescent quantum dots (Qdots) to label outflow pathways.
  • Employed immunofluorescence and RT-PCR for versican distribution analysis.
  • Utilized lentiviral shRNA to silence versican and CS GAG synthesis in anterior segment cultures.

  • Versican levels varied segmentally within the TM, inversely correlating with Qdot labeling.
