Background:

  • Ethanol exposure during critical brain development periods causes significant central nervous system (CNS) damage.
  • The precise cellular and molecular mechanisms driving these neurotoxic effects are not fully understood.
  • Endoplasmic reticulum (ER) stress, a cellular response to protein misfolding, is implicated in neurodegeneration.

Purpose of the Study:

  • To investigate whether ethanol exposure induces ER stress in the developing brain.
  • To identify the specific ER stress pathways activated by ethanol in immature neurons.

Main Methods:

  • Utilized a third-trimester equivalent mouse model for ethanol exposure.
  • Administered acute subcutaneous ethanol injections to 7-day-old mice.
  • Examined the expression of ER stress-inducible proteins (ERSIPs) and associated signaling pathways.

Main Results:

  • Ethanol exposure significantly upregulated ERSIPs and activated key ER stress signaling pathways, including ATF6, CHOP/GADD153, GRP78, and mesencephalic astrocyte-derived neurotrophic factor.
  • Phosphorylation of IRE1α, eIF2α, PERK, and PKR was observed, indicating pathway activation.
  • Ethanol-induced ER stress markers were detected in neurons within the cerebral cortex, hippocampus, and thalamus within 4 hours and persisted for over 24 hours.

  • This finding provides new insights into the detrimental effects of ethanol on the developing central nervous system.
