Summary
This study found no significant association between T-cell receptor beta (TCRB) gene polymorphism and endometriosis in an Italian population. Further research is needed to explore the role of TCRB in endometriosis pathogenesis.
Area of Science:
- Immunogenetics
- Reproductive Medicine
- Autoimmune Diseases
Background:
- Endometriosis is suspected to have an autoimmune etiology, supported by observed antibodies against the endometrium and high comorbidity with autoimmune and atopic diseases.
- Polymorphisms in T-cell receptor genes, such as T-cell receptor beta (TCRB), are relevant for studying immune responses and autoimmune conditions.
- The single nucleotide polymorphism (SNP) rs1800907 in TCRB has been previously investigated in autoimmune diseases.
Purpose of the Study:
- To investigate the association between T-cell receptor beta (TCRB) gene polymorphism and endometriosis.
- To compare the allelic frequency of the TCRB SNP rs1800907 between endometriosis patients and a control group.
Main Methods:
- A case-control study involving 70 patients with endometriosis and 120 controls.
- DNA analysis using polymerase chain reaction (PCR) followed by restriction digestion and sequencing to determine genotypes.
- Statistical analysis including Hardy-Weinberg equilibrium estimation and comparison of allele distributions using Pearson's and Fisher's tests.
Main Results:
- No significant difference in the allelic frequency of the TCRB SNP rs1800907 was found between endometriosis patients and controls (P = 0.87).
- The study had low statistical power (0.16) for detecting linkage disequilibrium.
Conclusions:
- The T-cell receptor beta (TCRB) gene polymorphism rs1800907 does not appear to be significantly associated with endometriosis in the studied Italian population.
- The findings suggest that TCRB may not play a major, evidential role in the pathogenesis of endometriosis within this cohort.
- Further investigation with larger sample sizes may be warranted to definitively exclude the involvement of TCRB in endometriosis.

Pharmacogenetics of Drug Targets: β₂-Adrenergic Receptors, Apo E, Thymidylate Synthase
Genetic polymorphisms in drug targets have emerged as critical determinants of interindividual variability in drug response and toxicity. Pharmacogenomic investigations increasingly focus on identifying these variations to personalize and optimize therapeutic interventions. A drug target may be a receptor, enzyme, or signaling protein involved in pharmacologic responses or disease-related pathways. While early pharmacogenetic studies focused primarily on drug metabolism, current research...
Diversity of Antigen Receptors
Antigen receptors are essential components of the immune system crucial in defending the body against foreign invaders. These receptors are present on the surface of B and T cells, enabling them to recognize antigens and mount an appropriate immune response.
Before encountering any antigen, lymphocytes express these receptors. On B cells, the antigen receptor is a membrane-bound antibody molecule called BCR; on T cells, it is a T cell receptor or TCR. B and T cell receptors are composed of two...

Receptor Downregulation in MVBs
Multivesicular bodies (MVBs) are mature endosomes that sort ubiquitinated proteins and then fuse with lysosomes to degrade the sorted proteins. Epidermal growth factor (EGF) and its receptor (EGFR) form a complex that can be internalized through endocytosis, sorted into an MVB, and later degraded.
The EGFR can initiate signaling pathways that lead to cell proliferation, migration, and differentiation. Overexpression of EGFR stimulates cells to proliferate. Excessive EGFR activation may...

T Cell Activation and Clonal Selection
T cells are integral to our adaptive immune system, recognizing and effectively responding to foreign antigens. T cell activation and clonal selection are pivotal in orchestrating this immune response. This article elucidates these mechanisms, detailing the roles of cluster of differentiation (CD) markers, major histocompatibility complex (MHC) molecules, costimulatory signals, and the process of clonal selection.
