Targeted Cancer Therapies02:57

Targeted Cancer Therapies

The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against specific...

Summary

Targeting CD30, a protein found on certain cancer cells like Hodgkin lymphoma and ALCL, shows promise. Antibody-drug conjugates targeting CD30, such as brentuximab vedotin, demonstrate significant efficacy with manageable toxicity in clinical trials.

Area of Science:

  • Oncology
  • Immunology
  • Molecular Biology

Background:

  • CD30 is selectively expressed on various lymphoid and non-lymphoid malignancies, including Hodgkin lymphoma and anaplastic large cell lymphoma (ALCL).
  • Restricted expression of CD30 on normal cells enables targeted therapy against malignant cells.
  • CD30, a member of the tumor necrosis factor (TNF)-receptor superfamily, possesses diverse biological functions.

Purpose of the Study:

  • To review the role of CD30 as a therapeutic target in CD30-expressing malignancies.
  • To highlight the potential of novel therapeutic strategies, particularly antibody-drug conjugates, for targeting CD30.

Main Methods:

  • Review of preclinical and clinical data on CD30-targeted therapies.
  • Analysis of the efficacy and safety profiles of antibody-drug conjugates, such as brentuximab vedotin.
  • Examination of the biological functions and signaling pathways associated with CD30.

Main Results:

  • Antibody-drug conjugates targeting CD30 have demonstrated significant clinical activity in early-phase trials.
  • Brentuximab vedotin has shown striking efficacy in patients with Hodgkin lymphoma and ALCL.
  • Therapies targeting CD30 exhibit manageable toxicity profiles.

Conclusions:

  • Targeting CD30 represents a promising therapeutic strategy for CD30-positive hematological and other malignancies.
  • Antibody-drug conjugates targeting CD30 offer a novel and effective treatment option.
  • Further investigation into CD30-targeted therapies is warranted for various CD30+ cancers.
