In vitro effect of actinomycin D on human neutrophil function
1Graduate Institute of Medical Sciences, National Defense Medical Center, Taipei, R.O.C.
Actinomycin D (ACT-D) significantly impairs human neutrophil functions, including bacterial killing and directed migration. This study reveals ACT-D
Area of Science:
- Immunology
- Cell Biology
- Pharmacology
Background:
- Neutrophils are critical immune cells for combating bacterial infections.
- Understanding the impact of pharmacological agents on neutrophil function is essential for clinical applications.
Purpose of the Study:
- To investigate the effects of Actinomycin D (ACT-D) on key human neutrophil functions.
- To assess ACT-D's impact on chemotaxis, oxidative burst, phagocytosis, and intracellular killing.
Main Methods:
- Human neutrophils were pretreated with varying concentrations of ACT-D.
- Neutrophil chemotaxis was assessed using fMLP as a chemoattractant.
- Chemiluminescence (CL), superoxide (O2-) production, phagocytosis, and intracellular killing of Salmonella enteritidis were measured.
Main Results:
- ACT-D (1-10 µg/ml) significantly reduced neutrophil chemotaxis towards fMLP.
- ACT-D pretreatment also depressed phorbol myristate acetate-induced CL and superoxide production.
- While phagocytic uptake was unaffected, ACT-D significantly impaired intracellular bacterial killing (P < 0.01).
Conclusions:
- Actinomycin D profoundly impairs human neutrophil-mediated intracellular bacterial killing, likely via effects on respiratory burst activity.
- ACT-D also significantly inhibits directed neutrophil migration (chemotaxis).
- Neutrophil viability remained high (>98%) even at tested ACT-D concentrations.
